Mortality, behaviour and morphological aberrations were observed at 24, 48, 72 and 96 hours.

Mortalities (96 hour) at 0, 550, 1090, 1640, 2180 or 2730 µg/L (nominal) were 0, 0, 10, 90, 100 or 100%, respectively. Treatment-related effects (96 hour) at 0, 550, 1090, 1640, 2180 or 2730 µg/L (nominal) were observed in 0/10, 0/10, 1/10, 10/10, 10/10 or 10/10 fish respectively. The 96 hour LC50 or EC50 of d-limonene to fathead minnow (Pimephales promelas) were calculated to be 720 (618-839) µg/L or 688 (606-782) µg/L, respectively.

A QSAR prediction was performed to assess the acute toxicity of test item d-limonene to fish. This QSAR has been validated to be compliant with the OECD recommendations for QSAR modeling (OECD, 2004) and predicts the endpoint value which would be expected when testing the substance under experimental conditions in a laboratory following Guideline for Testing of Chemicals No. 203, "Fish Acute Toxicity Test", referenced as Method C.1 of Commission Regulation No. 440/2008. The criterion was the LC50 (Median Effective Concentration), a statistically derived concentration which is expected to cause mortality in 50% of test animals within a period of 96 hours.

 

The QSAR determination of the acute toxicity to fish was based on validated data derived from 96-h LC50 tests on fish, for which the concentrations of the test item had been determined by chemical analyses over the test periods.

 

The 96-h LC50 (based on mortality) is predicted as 0.46 mg/L with 95 %-Confidence Limit between 0.40 and 0.52 mg/L.

 

This toxicity study is considered acceptable and satisfies the guideline requirements for acute fish toxicity study.

Based on the results of this study, d-limonene is classified dangerous for the environment according to Directive 67/548/EEC "R50 Very toxic to aquatic organisms" and hazardous to the aquatic environment according to Regulation (EC) No 1272/2008 (CLP) - Acute Category 1.
