Details on study design:
TEST SITE: Approximately 24 hours prior to application of the test substance, hair was removed with electic clippers from the dorso-lumbar region of each rabbit exposing an area of skin approximately 100 mm x 100 mm. Approximately 0.5 mL of the test substance was applied under a 25 mm x 25 mm gauze pad to one intact site on each animal. Each treatment site was covered with "Elastoplast" elastic adhesive dressing for four hours.

REMOVAL OF TEST SUBSTANCE: At the end of the exposure period, the semi-occlusive dressing and gauze pad were removed and the treatment site was washed with warm water (30-40°C) to remove any residual test substance. The treated area was blotted dry with absorbent paper.

SCORING SYSTEM:
Erythema and eschar formation: No erythema 0; very slight erythema (barely perceptible) 1; well-defined erythema 2; moderate to severe erythema 3; severe erythema (beef redness) to slight eschar formation (injuries in depth) preventing erythema reading 4.

For (2,4,6-trioxotriazine-1,3,5(2H,4H,6H)-triyl)tris(hexamethylene) isocyanate (EC 223-242-0, CAS 3779-63-3, mono constituent substance), in the following referred to as "HDI Trimer pure", a read across from the close structural analogue HDI oligomers, isocyanurate type (EC 931-274-8, CAS 28182-81-2, UVCB) is applied. A justification for the read-across with special focus on inhalation toxicity was elaborated and is attached to the chapter endpoint summary. Based on this justification all available toxicological data for HDI oligomers, isocyanurate type can be used for the toxicological evaluation of HDI Trimer pure. This approach is in accordance with Annex XI, section 1.5 of the REACH Regulation (EC) No 1907/2006.

Executive summary:

An Acute Dermal Irritation/Corrosion test was conducted according to OECD TG 404. In this study very slight erythema with or without very slight to slight oedema was seen in all animals (Mean scores of time points 24, 48, and 72 h: 1 for erythema, 0.8 for oedema). These reactions gradually ameliorated and had resolved in all instances by Day 5 or Day 6. No signs of toxicity or ill health were observed.

PROCEDURE: The eyes of each animal were examined prior to instillation of the test substance to ensure that there was no pre-existing corneal damage, iridial or conjunctival inflammation.
One animal was treated in advance of the others, to ensure that if a severe response was produced, no further animals would be exposed. Approximately 0.1 mL of the test substance was placed into the lower everted lid of one eye of each animal. The eyelids were gently held together for one second before releasing.

Additional information

Validated in vitro assays (OECD TG 431 and 439) revealed no corrosive and no irritant property to the skin. For investigating the ocular irritant property an in vitro human corneal epithelium (HCE) test, as prevalidated in 2004 is available. This model is recognized in the scientific community as a highly valuable model for the identification of substances that do not require classification for serious eye damage/eye irritancy (e.g. Cotovio et al., Toxicol. in Vitro, 24, 2010, 523 -537), and is routinely used by cosmetic and pharmaceutical companies. As test result the substance was concluded to be non-irritant to the eye.

In vivo Irritation/Corrosion studies with the read-across substance according to OECD TG 404 or 405 revealed slight skin and very slight eye irritation properties, respectively.

The read-across substance was proven to be respiratory irritating based on acute and repeated inhalation toxicity studies.

HDI Trimer pure itself was also found to be a pulmonary irritant based on a study according to the recommendations of TRGS 430 (Technical Rule for Hazardous Substances 430, published by the German Federal Ministry of Labour and Social Affairs, last update 2009; report AT06458, Bayer AG, 2012, see chapter Acute toxicity: inhalation). In this study clinical signs, lung weights and bronchoalveolar lavage analysis revealed findings suggestive of pulmonary irritation at concentrations higher than 15.7 mg/m³. The NO(A)EL of the study was determined to be 3.2 mg/m³.

Justification for classification or non-classification

According to Regulation (EC) No 1272/2008, Annex I, no classification is warranted for skin or eye irritation/corrosion.

Read across from a close structural analogue reveals only slight irritating effects to the skin and very slight irritating effects to the eye of rabbits, both not justifying classification. Additionally, (2,4,6-trioxotriazine-1,3,5(2H,4H,6H)-triyl)tris(hexamethylene) isocyanate

was tested in in vitro assays for skin corrosion and irritation and eye irritation. Also from the in vitro assays no corrosive or irritant property was concluded.

According to Regulation (EC) No 1272/2008, Annex I, classification as STOT SE 3 (H335: May cause respiratory irritation) is warranted.
