Description of key information

In the absence of specific data on the ADME of L-menthyl acetate, its physicochemical properties and relevant toxicity data (where available) were assessed for insights into likely ADME characteristics. L-menthyl acetate is a liquid with a MW of 198 g/mol, vapour pressure of 450 Pa at 25°C, water solubility of 21.0 – 25.4 mg/L and a logPow of 4.39. Due to its molecular weight below 500 g/mol and slight solubility in water, the absorption via the oral route is likely to be moderate. Absorption of L-menthyl acetate itself via the inhalation route will be low due to its molecular weight and low solubility. Low dermal absorption is expected. Based on a precautionary approach, an absorption default value of 100% is considered appropriate for oral and inhalation routes and 50% for the dermal route. L-menthyl acetate will be hydrolysed in mammals to menthol and acetic acid. Ester hydrolysis is catalysed by classes of enzymes recognized as carboxylesterases or acetylesterases. These enzymes occur in most mammalian tissues but predominatly in hepatocytes. The main elimination is thus considered to be via urine and bioaccumulation is unlikely.
