Determination of the test item
The samples were analyzed with a LC-MS/MS method, implemented under non-GLP and documented finally in the GLP raw data. The method will be validated.
The analytical method and the results of the method validation were added to the final report.

Sampling schedule
The limit loading and the control were analytically verified in the fresh media at the start of exposure and at one renewal interval (0 and 72 hours) and in the corresponding 24-hours aged media after 24 and 96 hours.

Limit concentration
A saturated solution prepared with 20 mg test item/L was tested as a threshold concentration in a limit test. The selection of the test concentration was based on the derivation of a threshold concentration (TC) from the data of the test item of results of a daphnia acute immobilization test (EC50 (0-48 h) = >saturated solution (nominal concentration of 20 mg/L) and of an alga toxicity test (ErC50 (0-72 h) = >saturated solution (nominal concentration of 20 mg/L).

Preparation of the saturated solution
A saturated solution with a nominal loading rate of 20 mg test item/L was prepared 24 ± 1 hour prior to the start of the exposure and the renewal. An appropriate amount of the test item was weighed out. The test item was inserted into a glass bottle with an appropriate amount of dilution water. The saturated solution was stirred for 24 ± 1 hours (1100 rpm, room temperature) with a magnetic stirrer. After the stirring period undissolved particles were removed by membrane filtration (membrane filter 0.45 µm, RC, Macherey-Nagel). The filter was saturated in order to avoid adsorption during the filtration. The first 25 mL of the filtrate were discarded. The filtration was interrupted for 15 minutes to allow adsorption and saturation of the filter material with dissolved test item. Thereafter, the filtration was continued. The next 25 mL was discarded. The following filtrate, i.e. the saturated solution, was used in the test. During filtration, the filter was always kept covered. The saturated solution was checked via laser beam (Tyndall effect) for undissolved test item. No presence of undissolved test item during the test was observed.

Control
7 fish in dilution water (without test item but from the same source and age as used for the test solution) were tested under the same test conditions as the test replicate.

TEST ORGANISM

- Source: All fish used in the test were gained at NOACK LABORATORIEN from a single brood stock. (supplier: Umweltbundesamt, Schichauweg 58, D-12307 Berlin)

- Method of breeding:Holding was performed at the test facility at 23 +/- 2 °C and diffuse light (540 – 1000 Lux, photoperiod of 12 to 16 h light / 12 to 8 h dark). The water was changed at least once per week. The dissolved oxygen concentration was more than 80 % of the air saturation value. Food was provided 3 times per week. The amount of food was 4 % of the fish body weight per feeding day. The test fish were not fed 24 h before the test started. Zebrafish with at least 9 days of acclimatization and mortality < 5% within these days prior to the start of the exposure were used in the test. No mortality was observed during this time. No disease treatments were administered throughout holding and testing.

measured in all test vessels at the start and the end of exposure as well as daily and after renewal of the test media from fresh and old test media.
Chlorine and nitrate were measured from each batch of dilution water used in the test.
Total hardness and TOC were determined at the start of the exposure in the dilution water. During the test, the water temperature was recorded continuously (once per hour) with a data logger. The light intensity on the surface of the test aquaria was measured at the start of the exposure. The solution appearance and behavior was observed daily before and after renewal.

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) : Fish were considered dead if there was no visible movement (e.g. gill covers movement) and if touching of the caudal peduncle produced no reaction. Records were kept of visible abnormalities (e.g. loss of equilibrium, swimming behaviour, respiratory function, pigmentation, etc.). Observations were made after 2 ± 0.5 h, 5 ± 1 h and 24 ± 2 h after the start of the exposure. On days 1-4 of the test, all vessels with living fish were inspected twice per day.

The effect level is given based on the geometric mean measured test item concentration of Exolit 775. The threshold concentration of the test item at the nominal loading rate 20.0 mg/L corresponding to the geometric mean measured concentration of 2.48 mg/L was found to have neither lethal nor non lethal effects on zebrafish.
In conclusion, the LC0 corresponds to the geometric mean measured concentration of ≥ 2.48 mg test item/L.





Executive summary:

 


The acute toxicity of the test item  to fish (zebrafish) was determined according to the principles of the OECD-Guideline for Testing of Chemicals No. 203 (2019) from 2020-10-04 to 2020-10-09 with the definitive exposure period from 2020-10-05 to 2020-10-09 at the test facility.


 


The test item is a white solid. A saturated solution prepared with 20 mg test item/L was tested as a threshold concentration in a limit test. The selection of the test concentration was based on the derivation of a threshold concentration (TC) from the data of the test item of results of a daphnia acute immobilization test (EC50 (0-48 h) = >saturated solution (nominal concentration of 20 mg/L) and of an alga toxicity test (ErC50 (0-72 h) = >saturated solution (nominal concentration of 20 mg/L).


 


The study was performed over a period of 96 hours under semi-static conditions with daily renewal of the test solutions. Seven test organisms were exposed to the test item concentration and the control, respectively. Water quality parameters (temperature, pH-value and oxygen-saturation) measured at 0, 24, 48, 72 and 96 hours were determined to be within the acceptable limits.


The test solutions were visually clear throughout the study.


 


The loading rate of the test item  was analytically verified via LC-MS/MS in fresh media at the start of the exposure and at one renewal interval (0 hours and 72 hours) and in the corresponding 24-hours aged media after 24 hours and 96 hours in the limit test item loading and the control.


The measured concentrations of the test item were in the range of 2.34 to 2.43 mg/L of the nominal concentration in the fresh media at start of the exposure intervals. At the end of the exposure intervals the test item concentration were in the range of 117% to 101% of the initially measured test item concentration.


All effect values given were based on the geometric mean measured concentration of the test item.


 


 


All validity criteria of the test guideline were met. The threshold concentration of the test item at the nominal loading rate of 20.0 mg/L corresponding to the geometric mean measured concentration of 2.48 mg/L was found to have neither lethal nor non lethal effects on zebrafish.
