PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method:
The substance was prepared with addition of Tween 80 and dilution water. The stock solution contains 500 mg/l of the test substance and 100 mg/l of the dispersant. The stock solution was stirred 10 minutes. Following this, serial dilutions of this test medium were prepared in order to obtain the desired test concentrations. Additionally a culture medium and solvent control was tested in parallel.

Additional information

All substances in this category share a similar structure with a molecular weight > 500 g/mol. Based on the high molecular weight and insolubility, the substances in this category will not be able to penetrate biological membranes. Thus, no systemic exposure is expected. This is confirmed by the environmental studies on aquatic invertebrates for two substances in this category: CAS 1328-53-6 and CAS 147-14-8. The environmental toxicity data can be used for read-across to all substances in this category.


In all studies, Daphnia magna was used as the test organism, and no analytical measurements were performed. Two studies on substance CAS 1328-53-6 are available. In the study conducted according to EPA-660/3-75-009, the 48-h EC50 for mobility was determined to be 154 mg/L (Sun Chemicals 1978, K2). The second study was conducted according to EU Method C2. The 48-h EC50 for mobility was determined to be > 500 mg/L (BASF AG 1988, K2).  In the study performed with substance CAS 147-14-8 according to OECD TG 202, the 48-h EC50 for mobility was determined to be > 500 mg/L (BASF AG 1989, K2). 


The determined LC50 values were significantly higher than the water solubility of the test substance. Based on the available information, it is concluded that all substances in this category are not acutely toxic to aquatic invertebrates, with an LC50 of 154 mg/L.
