Description of key information

In vivo data for skin irritancy indicate moderate irritation potential (Anon 1970, OECD SIDS 1998). No in vitro assessment of skin irritancy is considered necessary.
An in vivo assessment of conjunctival reactions in rabbits indicated that chloroprene elicited conjunctivitis with recovery within ten days after instillation (OECD SIDS 1998). Evdience in humans and animal studies indicates that chloroprene may cause respiratory irritation.

Additional information

There is limited information on the skin and eye irritation potential of chloroprene (Anon 1970, OECD SIDS 1998). Studies in rabbits indicate that both dermal and ocular responses are likely following exposure to chloroprene. Studies have not shown chloroprene to be corrosive to the skin. An in vivo assessment of conjuctival reactions in rabbits indicated that chloroprene elicited conjunctivitis which lasted for 10 days.

The OECD SIDS (1998) for chloroprene reports that the primary effects of acute exposure to high concentrations in air in humans are irritation of the skin and mucous memrbranes, CNS depression and respiratory difficulties - no further details are provided.

In repeated dose inhalation studies conducted by NTP (NTP, 1998), hypoactivity, unsteady posture and rapid, shallow breathing occurred after a single exposure and bloody noses were observed after the second day of exposure. Bronchial and bronchiolar degeneration have been seen in rodents 2 days following exposures at 19.58 mg/l.

Justification for classification or non-classification

According to Annex VI to Regulation (EC) No 1272/2008, chloroprene is classified for irritancy hazard as: Eye Irrit. (H319) – Causes serious eye irritation, STOT SE 3 (H335) – May cause respiratory irritation and Skin Irrit (H315) – Causes skin irritation.

Based on the evaluation of available data, the classification of chloroprene for skin, eye and respiratory irrtiation is warranted.
