Description of key information

The acute toxicity of the surrogate test substance to Daphnia magna was assessed according to DIN 38412 L11 (BASF 1990). The 48 h-EC50 value was determined to be nominal 255 mg test item/L. In addition study results are available for all the other category members, all revealing EC50 values well above 100 mg/L.

Additional information

The acute toxicity of the test substance 3-methylbutan-1-ol to Daphnia magna after 48 hours was assessed in a static test according to DIN 38412 L11 (BASF 1990). The nominal concentrations of the test substance were 0, 58, 100, 180, 320, and 580 mg/L. No concentrations of the test substance were measured. The number of Daphnids per vessel was 5 and the number of vessels per concentration was 4. A positive control with the reference substance potassium dichromate was performed. The 48h-EC50 of the reference substance potassium dichromate was determined to be 1.45 mg/L. The validity criteria of the test were met. The 48h-EC50 value of the test substance was determined to be nominal 255 mg test item/L.

Supplementary the acute toxicity of the read across substance pentan-1-ol to Daphnia magna was assessed. The mobility of the Daphnia was determined in a static 48-hour test by visual observation after 3, 6, 24 and 48 hours. The 48h-EC50 value was determined to be nominal 341 mg test item/L.

 

In addition, the acute toxicity of the surrogate test substance 2-methylbutan-1-ol on aquatic invertebrates was investigated under GLP conditions according to OECD 202 (BASF 2011). The 48h-EC50 value was determined to be greater than 173 mg/L (value based on measured concentration).

 

Study results are also available for the fourth category member pentanol, branchend and linear. The EC50 value of this substance was determined to be greater than 120 mg/L in a GLP guideline study with Daphnia magna according to OECD 202 (BASF 2011).

 

In conclusion, the 48h-EC50 value of the test substance and of the read across category members was determined to be greater than 100 mg/L. The EC50 value determined with the test substance 3-methylbutan-1-ol was chosen as “key value” and used for PNEC derivation.
