Skin sensitisation

Endpoint conclusion
Endpoint conclusion:
adverse effect observed (sensitising)
Additional information:

No guideline-conform, reliable study on skin sensitisation of o-Chlorobenzylchloride has been identified.

There is one study report available with low reliability (Landsteiner and Jacobs, 1936). The study was conducted in guinea pigs, suggesting that eight out of thirteen animals tested gave a positive response to o-Chlorobenzylchloride. Rao et al.(1981) evaluated various chemical groups for their skin sensitization potential in the guinea pig. Positive responses occurred with benzyl chlorides. In the Guinea Pig Maximization Test according to OECD guideline 406 and GLP the position isomer p‑Chlorobenzylchloride was a sensitizer (Hoechst AG, 1986).

Based on this weight of evidence, o-Chlorobenzylchloride is considered to be a sensitising compound.

Migrated from Short description of key information:
o-Chlorobenzylchloride is considered to be a dermally sensitisizing agent.

Justification for classification or non-classification

Based on the available data, o-Chlorobenzylchloride is classified as dermally sensitisizing according to EU Directive 67/548/EC and EU Regulation No.1272/2008 and CLP:

R43, may cause sensitisation by skin contact (according to Directive 67/548/EEC)

Skin Sens. cat. 1, H317, may cause allergic skin reaction (according to Regulation 1272/2008/EC)
