- Sampling method: All concentration levels and the control were analytically verified via GC-MS/MS after 0 hours (start of exposure) and 72 hours (end of exposure). Separate replicates for the test item analysis at the beginning of the exposure were prepared without algae. For the test item analysis after 72 hours, separate replicates of the concentration levels 1.0 to 10.0 mg/L and control were prepared and incubated under test conditions. Samples of the concentration levels 31.6 to 100 mg/L were taken out of the test replicates. No specific treatment for removal of algae cells from the samples was applied.

PREPARATION AND APPLICATION OF TEST SOLUTION:
A stock solution of 100 mg/L was freshly prepared with dilution water. Direct addition of the test item and shaking until the test item was completely dissolved.

EFFECT PARAMETERS MEASURED:
- Chlorophyll measurement: The cell density was measured daily via Chlorophyll a-fiuorescence, excitation at 436 nm, emission at 685 nm. Dilution water was used as a background signal. No self-fluorescence was found in the preliminary range finding test at the concentration level of 100 mg/L.
- Microscopic evaluation: Microscopic evaluation of the cells at the start and the end of exposure was carried out. The ceils were checked for unusual cell shapes, colour differences, differences in chloroplast morphology, flocculation, adherence of algae to test containers and agglutination of algae cells.

Reported statistics and error estimates:
The NOEC and LOEC were determined by calculation of statistically significant differences of growth rates and yield. As a standard, One Way Analysis of Variance (ANOVA) and Dunnett's test were used for NOEC/LOEC calculations. When running a One Way Analysis of Variance, a Normality test and an Equal Variance test were done first. P-values for both Normality and Equal Variance tests are 0.05. The α-value (acceptable probability of incorrectly concluding that there is a difference) is α=0.05.
The data for the tables in this report were computer-generated and have been rounded for presentation from the full derived data. Consequently, if calculated manually based on the given data minor deviations may occur from these figures.

Measured Exposure Concentrations during the Definitive Test

The concentrations of the test itm were determined in the fresh media (0 h) and old media (72 h) of all tested concentration levels and the control via GC-MS/MS. For results, see Table 2. The measured concentrations of the test item in the fresh media (0 h) were in the range of 82 to 97 % of the nominal values at all tested concentration levels. The measured concentrations in the old media (72 h) were in the range of 9 to 94 % of the nominal values of all concentrations. Effects were found only at the highest concentration of 100 mg/L. Therefore, the nominal concentrations were used for calculations.

The concentrations of the test item in all test concentrations and the control were analytically verified by GC-MS/MS at the start and the end of the exposure. The measured concentrations of the test item in the fresh media (0 hours) were in the range of 82 to 97 % of the nominal values at all tested concentration levels. The measured concentrations in the old media (72 hours) were in the range of 81 to 94 % of the nominal values of the two highest concentrations (31.6 and 100 mg/L) and in the range of 9 to 16 % of the concentration levels 1.00 to 10.0 mg/L.
