Two valid studies, one following guideline methods and the second conducted according to a non-guideline method, are available.

The 96-h LC50 values of the two acute fish toxicity tests with two different species ranged from ca. 87 mg/L in Leuciscus idus to 720 mg/L in a test on Pimphales promelas embryos. In the static test on the toxicity of caffeine to Leuciscus idus which was conducted according to the German standard guideline DIN 38412 part 15, fish (n = 10 per replicate) were exposed for 96 hours to six concentrations of caffeine ranging between 10 - 464 mg/L (1 replicate) and to control water (1 replicate). The 96 hour LC50 of 87 mg/L is based on the nominal test concentrations.

In a 120-hour static test, the toxicity of caffeine to embryos of Pimphales promelas (n = 10 per replicate) and the malformation of embryos due the exposure to caffeine was determined. Groups of 30 - 40 embryos were tested. The LC50 after 120 hours was 720 mg/L, whereas the EC50 for malformation was determined to be 70 mg/L (LOEC = 20 mg/L).
