Analytical evaluation of the concentrations of Isobornyl acrylate and the control were carried out via GC-FID from freshly prepared media after 0 and 72 h, and from 24 h old test media after 24 and 96 h.

All fish used in the test were gained at Dr.Noack-Laboratorien, from a single brood stock (supplier: Umweltbundesamt, Schichauweg 58, D-12307 Berlin).

Holding:
Fish were held at the test facility at 23  2 °C and diffuse light (0.1-10 µmol photons • m-2 • s-1, natural photoperiod).
The water was changed at least once per week.
The dissolved oxygen concentration was more than 80 % of the air saturation value.

Zebrafish with at least 12 days of acclimatisation and mortality < 5 % within this period of time were used in the test. No disease treatments were administered throughout holding and testing.

Food (Sera Vipan; Sera GmbH, D-52518 Heinsberg) was provided 3 times per week. The amount of food was 4 % of the fish body weight per feeding day.
The test fish were not fed 24 h before the test started.

Dilution water was prepared according to the test guideline (OECD 203) following internal SOP’s.

Test concentrations:
Aliquots of the stock solution were shaken for 24 h, then they were diluted with purified drinking water (dilution water).


Test priciples and procedure:

10 fish were used for each test concentration including controls.
Only fish in good health and free from any apparent malformation were used.


15-l glass aquaria were used as test vessels and filled with 10 l of test solution. The test animals were added to the previously prepared test solution; controls were kept in dilution water (blank).

The aquaria were slightly aerated via a glass capillary throughout the test.


The fish were exposed to various concentrations of the test substance for a period of 96 hours under semi static conditions. The test medium was changed completely every day. Visible abnormalities observed during the first 6 hours of the experiment were recorded. Mortalities and visual abnormalities were recorded at 24 h, 48 h, 72 h, and 96 h.

The test temperature was 21.8° C to 22.1 ° C. The pH of the dilution water was 7.8 to 8.1. The oxygen saturation in all test vessels was between 86 % and 100 %.
The light/dark rhythm was 14h/10h.

The pH, temperature and the oxygen concentrations of the control and of all the test concentrations were measured daily.

In a valid guideline study (OECD 203, semi-static conditions), the 96 hr LC50 for Danio rerio (Teleostei, Cyprinidae) is 0.704 mg/L .
Executive summary:

 In a valid 96-hr acute toxicity study according to OECD TG 203 with GLP, Danio rerio were exposed to Isobornyl acrylate at 0 (control), 0.155 – 0.341 – 0.751 – 1.65 – 3.64 – 8.00 mg/L nominal and 0 (control), 0.073 – 0.159 – 0.431 – 1.15 – 2.63 – 6.28 mg/L measured concentrations under semi-static conditions.

The 96-h LC50was 0.704 (0.431 – 1.15 95 % confidence limits) mg a.i./L based on mortality.

The LC100and NOEC values, based on mortality effects, were 1.15 mg/L and 0.431 mg a.i./L, respectively. The effect concentrations refer to the geometric mean measured concentrations.
