Hydrolysis

A hydrolysis study was performed according to OECD Guideline 111. The major constituents of the test item (1H-Benzotriazole-1-methanamine, N,N-bis(2-ethylhexyl)-5-methyl-, 1H-Benzotriazole-1-methanamine, N,N-bis(2-ethylhexyl)-4-methyl- and 1H-Benzotriazole-1-methanamine, N,N-bis(2-ethylhexyl)-6-methyl-) hydrolyse directly and quantitatively when it is solved in the buffer solutions. Thus the test item is not considered to be hydrolytically stable at pH 4, 7 and 9. The determination of the hydrolysis rate is not possible because of the very fast hydrolysis. Methyl-1H-benzotriazole (tolyltriazole) was detected as hydrolysis product. Additionally the following transformation products were proposed: Formaldehyde (CAS No. 50-00-0), Diethylhexylamine (CAS No. 106-20-7)

Biodegradation

The ready biodegradability of the test item was assessed over a 28 day period by the modified Sturm test (OECD 301B). The nominal test concentrations were 10 and 20 mg a.s./L. The test material attained up to 11 and 7 % biodegradation after 28 days whereas the positive control substance (Aniline) was 94.4 % degraded 28 days. These results indicate that the test item cannot be classified readily biodegradable under conditions of the test.

The inherent biodegradability of the test item was assessed over a 28 day period by the modified Zahn-Wellens Test (OECD 302B). The nominal test concentrations was 10 mg a.s./L. The test material attained up to 60 % biodegradation after 28 days whereas the positive control substance was 100 % degraded after 27 hours. These results indicate that the test substance is inherently biodegradable in this test when measured by specific analysis.

The constituents and proposed hydrolysis products are moderately and readily biodegradable (bisethylhexylamine and formaldehyde) or not readily biodegradable (tolyltriazole).

Bioaccumulation

The multiconstituent substance is not readily biodegradable but the main constituents hydrolyse fast. The log Pow of the remaining major constituents and proposed degradation products formaldehyde (CAS 50-00-0, logPow 0.35) and tolyltriazole (CAS 29385-43-1, logPow 1.71) is below 3. Based on weights of evidence with QSAR BCF data, it can be concluded that bisethylhexylamine (CAS 106-20-7, logPow 6.56 for the uncharged molecule and logPow 7.3 at pH 7.5) does not significantly accumulate in organisms.

However, the BCF for the main component of the multiconstituent substance was calculated to be approx. 166 (considering biotransformation) - 1680 L/kg ww (without considering metabolism) using EPI Win (BASF SE, 2013).

Secondary poisoning is therefore, highly unlikely to occur in any extent for the test item.

Adsorption / Desorption

The main constituents of the substance hydrolyse fast. The remaining major constituents and proposed degradation products are formaldehyde (CAS 50-00-0, logPow 0.35), bisethylhexylamine (CAS 106-20-7, logPow 6.56 for the uncharged molecule and logPow 7.3 at pH7.5) and tolyltriazole (CAS 29385-43-1, logPow 1.71).

However, the KOC for the main component of the multiconstituent substance was calculated to be approx. 140400-721000 L/kg using EPI Win (BASF SE, 2013). Based upon these data, significant adsorption to soil is only expected for bisethylhexylamine (KOCWIN v2.00).

Assessment of the degradation products and constituents:

The following transformation products were proposed: Formaldehyde (CAS No. 50-00-0), Bis ethyhexylamine (CAS No. 106-20-7). Tolyltriazole (CAS 29385 -43 -1) is detected as degradation product and additionally a major constituent of the uvcb substance.

 

Formaldehyde is registered under REACH Regulation (EC) No 1907/2006. According to the disseminated registration data the substance is well water soluble, readily biodegradable, volatile and mobile in soil. The BCF was determined to be 0.396 (logPow = 0.35), thus it was not classified as PBT and vPvB. In addition, Formaldehyde has not be classified as environmental toxic according to Regulation (EC) No 1272/2008 (CLP/GHS).

 

Bis(2-ethylhexyl)amine is registered under REACH Regulation (EC) No 1907/2006. According to the registration data the substance has a low water solubility is not readily biodegradable but moderately biodegradable. Based on a weight-of-evidence approach, the substance is regarded to be not persistent.

The pKa of 10.6 indicates that the molecule will exist primarily as a cation in the environment and cations generally adsorb stronger to soils containing organic carbon and clay than their neutral counterparts.

The logPow was determined to be 7.3 (at 25 °C, pH 7.5) and 6.56 for the uncharged molecule. Based on weights of evidence with QSAR data, it can be concluded that the substance does not significantly accumulate in organisms. Thus in accordance with column 2 of REACH Annex IX no bioaccumulation study was performed. Therefore, the substance is not classified as PBT or vPvB. The substance is classified as chronic aquatic cat. 1, M-factor 1 (H410: Very toxic to aquatic life with long lasting effects.) according to Regulation (EC) No 1272/2008 (CLP/GHS).

Relevant data from registration dossier: Molecular weight 241.46 g/mol, Vapour pressure 0.0023 hPa, Water solubility 14 mg/L, logKow 7.3 (pH 7.5), pka 10.59, Henry's law constant 30.0229Pa3m3/mol corrected for pH 7, KOC 42354 pH7 (estimate according to Franco & Trapp (2008) using a logKow offor the uncharged molecule of 6.56 and a pka of 10.59 as input parameter), Biodegradability for exposure assessment: not biodegradable.

Tolyltriazole is subject to REACH registration (CAS 29385 -43 -1) according to Regulation (EC) No 1907/2006. However, data are not yet available. Also, no data on biodegradability are available so far and the estimated BCF-factor using EPIWIN was calculated to be 3.8 to 6.4 (logPow = 1.71, calculated using Episuite). Thus, the compound is not classified as PBT and vPvB. No data on tolyltriazole are available but QSAR estimations indicate that this substance is well water soluble, is probably not readily biodegradable, not bioaccumulating and mobile in soil.
