In the key study (Butler, 2013), the water solubility of the test material was investigated in a GLP study which was conducted in accordance with the standardised guidelines OECD 105 and EU Method A.6 using the flask method.

The preliminary water solubility test indicated that the column elution method should have been performed as the solubility was less than 0.01 g/L. However, due to the physical nature of the test material, it was not possible to use this method.

During method development, difficulties were encountered with traditional chromatographic techniques using HPLC columns. It was considered that the test material interacted with the columns in a manner that it was not possible to achieve consistent and reliable chromatography. Mass spectroscopy was considered to be a suitable means of detection.

Mixtures of test material and purified water were added to three separate flasks; 0.1028, 0.1019 and 0.1052 g were added to 100 mL of purified water for samples 1, 2 and 3, respectively.

After addition of purified water to the flasks, they were shaken at approximately 30 °C for 24 hours (sample 1), 48 hours (sample 2) and 72 hours (sample 3). After standing at 20 °C for a period of 24 hours, the contents of the flasks were filtered through 0.45 μm filters prior to dilution for analysis.

Under the conditions of this study, the water solubility of the test material has been determined to be less than or equal to 0.000156 g/L of solution at 20.0 ± 0.5 °C and is considered to be slightly soluble.

 

Supporting information is provided in the form of a review article (Summer, 2003) in which the water solubility of the test material was reported to be < 1 mg/L. No information is presented on materials and methods.
