PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Stock solution I was prepared by dissolving 100 mg test substance in tetrahydrofuran to a final volume of 10 mL. Stock solution II was obtained by taking 1000 µL of stock solution I and adding tetrahydrofuran to a final volume of 10 mL. Stock solution III was prepared individually for each test concentration. 80, 140, 250, 450 and 800 µL, respectively, of stock solution II was taken, and tetrahydrofuran was added to a volume of 10 mL. Between all dilution steps, the solution was mixed by inversion. For the test concentrations, 0.11 mL of the stock solution III was taken and dilution water was added to a final volume of 1.1 L.

One study is available on the short-term toxicity of 3,3,4,4,5,5,6,6,7,7,8,8,8-tridecafluorooctyl 2-chloropropenoic acid ester (CAS 96383-55-0) to aquatic invertebrates. The study was conducted according to the OECD guideline 202 and GLP (Matsumoto, 2012). As the test substance is poorly soluble in water, tetrahydrofuran was used as a solvent for the preparation of the test solutions. The test organism Daphnia magna was exposed under semi-static conditions to nominal test concentrations of 0.00080, 0.0014, 0.0025, 0.0045 and 0.0080 mg/L for 48 hours. At the end of the exposure period, 10% immobilization was observed only at the highest test concentration. The EC50 (48h) was therefore determined to be above the highest test concentration, corresponding to a measured value of > 0.00371 mg/L.
