- The determination of the actual test item concentrations was based on the measurement of the sum of all components of the test item. For this, duplicate samples were taken from each test concentration and the control at the start and at the end of the two 24-hour test medium renewal periods.
The concentrations of JASMONYL LG were analyzed in one of the duplicate test medium samples from the nominal concentrations of 32, 56 and 100 mg/L from all sampling dates. The samples of the nominal test concentrations of 10 and 18 mg/L were not analyzed since these concentrations were below the 48 hour NOEC determined in this test and, thus, were not relevant for the interpretation of the biological results.

- Reconstituted test water (ISO Test water) according to the OECD Guideline No. 202 was used in the study. It consisted of analytical grade salts dissolved in purified water.
Since the test item was determined to be volatile, the test was performed in glass vessels completely filled (without headspace) with 60 mL test medium and tightly sealed with glass stoppers to avoid losses of the volatile substance by evaporation (closed system).

- The study was performed with young daphnids of the species Daphnia magna Straus. A clone of this species (originally from the Daphnia Collection of the University of Basel/Switzerland in 2015) is successfully bred in IES Ltd Laboratories.
At the start of the test, the organisms used in the test were 6 to 24 hours old and were not first brood progeny.

- Test temperature:
- The test was performed in a temperature-controlled room with continuous monitoring of the room temperature. The water temperature was in the range of 21-22 °C.

- At the beginning and end of the test medium renewal periods, the pH values of the test media were between 7.6 and 7.9.

After 24 hours, the test organisms were transferred by wide-bore pipette in to clean test vessels with freshly prepared test medium of the corresponding concentration.

- The measured concentrations of the test item in the analytically analyzed test media of nominal 32 to 100 mg/L were between 87 and 102% of the nominal values at the start of the two test medium renewal periods. Thus, the correct dosage of the test item was confirmed. At the end of the test medium renewal periods, the chromatographic profile of the test item was not significantly changed and represented 84 to 98% of the nominal values (sum of peaks). The analytical results showed that the test design chosen (closed system and semi-static design) was appropriate for a sufficiently constant test item exposure of the daphnids. Therefore, the biological results are based on the nominal concentrations of the test item.
