- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Direct dissolution of test item in culture medium to prepare stock; serial dilution of stock to prepare test media. An amount of test item (5.50 g) was dispersed in 11 litres of reconstituted water and stirred
with a propeller stirrer (approximately 2000 rpm) for a period of 24 hours at 25°C. After 24 hours stirring the mixture was cooled to approximately 21 °C prior to filtration (0.2 microns) through a Suporcap (first 1 - 2 litres discarded to precondition the filter) to give a saturated solution of approximately 11 mg/L (based on the results obtained for the acute toxicity to Daphnia magna test (cited in full study report). An aliquot of the saturated solution (200 mL) was dispersed in a final volume of 2 litres of reconstituted water to give a nominal 1.1 mg/L stock solution. Aliquots of this nominal 1.1 mg/L stock solution (7.2, 23.6, 72.8, 236 and 728 ml) were each separately dispersed in a final volume of 2 litres of reconstituted water to give nominal test concentrations of0.0040, 0.013, 0.040, 0.13 and 0.40 mg/L. The concentration and stability of the test item in the test preparations were verified by chemical analysis on Days 0, 3, 7, 10, 14, 17 and 20 (fresh media) and on Days I, 4, 8, 11, 15, 18 and 21 (old media). Analysis of the freshly prepared test solutions throughout the test showed the measured test concentrations to range from 0.00344 mg/L to 0.427 mg/L throughout the test (equivalent to a range of 69% to I 07% of the expected nominal values). The majority of samples were observed to be within the accepted range of between 80% to 120% of nominal.

- Observations on body length and weight: In P1 generation: There was a significant effect on size and colour of the daphnids in that all the surviving daphnids at 0.40 mg/1 were observed to be markedly smaller and paler in colour than the control animals from Day 4 until 100% mortalities were observed on Day 11. surviving daphnids at 0.13 mg/L were also observed to be paler than the control animals on Days 9 and 11. On Day 12 100% immobilisation was observed at this test concentration. At 0.040 mg/L one out of four daphnids was observed to be small and pale on Day 12 and one out of two daphnids was observed to be pale when compared to the control animals on Days 15 and 16. The daphnids at the remaining test concentrations were observed to be the same size and colour as the control animals. In F1 generation: there was no noted differences to controls.

Based on the results of an acute toxicity test, Daphnia magna were exposed (10 replicates of a single daphnid per group) to an aqueous solution of the test item over a range of nominal test concentrations of0 (control), 0.0040, 0.013, 0.040, 0.1300, 0.40 mg/Lfor a period of 21 days. The saturated solution was prepared by stirring an excess of test material in test medium for a period of 24 hours prior to removal of the excess test material by filtration (0.2 µm). Test concentrations were prepared by dilution of a saturated solution of the test item. The saturated solution was prepared by stirring an excess of test item in test medium for a period of 48 hours prior to removal of the excess test item by filtration (0.2 µm). The test solutions were renewed daily (or > 3 times per week). Daphnids were fed daily with algal suspension. The numbers of live and dead adult (P1) daphnids and (F1) young daphnids was determined daily. It was considered validity criteria of the test guideline were fulfilled. Analysis of the saturated solution used to prepare the test concentrations throughout the test showed the measured test concentrations to range from 9.1 mg/L to 10 mg/L (82% to 94% of the expected nominal value). These results were consistent with the results from the pre-test preparation trials (11 mg/L) and the water solubility value of 10.1 mg/L. Analysis of the freshly prepared test solutions throughout the test showed the measured test concentrations to range from 0.00344 mg/L to 0.427 mg/L throughout the test (equivalent to a range of 69% to 107% of the expected nominal values). On this basis it was considered appropriate to base effect level estimates on time weighted mean measured concentrations. The test item 21-d EC50 (reproduction) was between 0.0039 and 0.012 mg/L based on time weighted mean measured concentrations. There was no significant difference (P ≥ 0.05) in terms of the number of young produced per adult when compared to the control and that the adult daphnids at 0.0039 mg/L were eliminated from the test by Day 21 due to a prolonged toxic effect of the test item.
