Biotransofrmation and kinetics

A study was conducted to determine the metabolic fate of NPE-2 in rainbow trout with a comparative in vivo/in vitro approach. The test item was administered in fish (p.o.) and urine was collected during 72 h. The fish were sacrificed, the gall bladder was excised and radioactivity was measured in tissues, viscera and carcasses. Metabolic profiles were performed by radio-HPLC and when possible metabolites were identified by LC/MS. For comparison, the biotransformation of the test item was also investigated in freshly isolated hepatocytes. Under the above test conditions, the major in vivo metabolite of NPE-2 was found to be the NPE-2 glucuronide and NP and/or NP glucuronide were not detected in any bile or hepatocyte sample . Furthermore NPE-2 was found to be widely spread in the fish body, however, it was not detected in fat (Cravedi J.P, 2001).
