Estimated value

The hydrosolubility was first estimated using 20 µL of pure test substance in 10 mL of water in a 15 mL capacity glass tube. The tube was firmly shaken and then allowed to stand for 1 hour so that the phases separated. A 5 mL was then sampled from the water phase and extracted in 2 mL chloroform as validated.

The concentration was measured as 409 mg/L, so the main test was performed according to the flask method.

Measured value

8 tubes were rotated for 4 days at 20 +/-1°C. Four tubes were sampled at 24 h intervals and checked for that no particles may affect the results.

The water solubility was calculated as mean from days 1, 2 and 3 which fulfilled the validity criteria: the standard deviation (6.4 mg/L) within the three successive values (287, 256 and 274 mg/L, rounded values) represented 2.4% of mean value.

pH of the water phase was 6.33 at the end of the test.

The equilibrium in water was achieved within 1 day, and the deletion of the incubation of the solutions at 30°C was justified.

Executive summary:

The water solubility of the test substance was determined under GLP based on EU A.6 guideline, flask method.

After preliminary testing, water was equilibrated with about 5 times the estimated substance amount, for 4 days, at 20°C. Concentrations were measured each day using a GC-FID validated method. Individual results fulfilled the validity criteria for reproducibility.

The water solubility of the test item was determined as 272 mg/L (at 20°C and pH=6.33).
