Sampling and analysis

Analytical monitoring:
no

Test solutions

Vehicle:
yes
Details on test solutions:
Stock solutions of 2,3-epoxypropyl neodecanoate were prepared in Analar acetone to yield final nominal concentrations of 1, 2, 5, 10, 20 and 50 mg/L when added to the treatment vessels. The final concentration of acetone was 0.1 ml/L.

Test organisms

Test organisms (species):
Oncorhynchus mykiss (previous name: Salmo gairdneri)
Details on test organisms:
Healthy S. gairdneri (rainbow trout) fingerlings were obtained from Itchen Valley Trout Farm, Airesford, Hampshire and acclimated to the test conditions for more than 10 days before use. A sample of ten of the fish used in the test had a mean length of 5.0 cm (range 4.4 to 5.9 cm) and a mean weight of .1.5 gm (range 0.9 to 2.3 gm).

Study design

Test type:
semi-static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
96 h
Post exposure observation period:
None.

Test conditions

Hardness:
Total hardness of the water was 250—270 mg/L as CaCO3. and the concentration of dissolved oxygen was 10.2—10.6 tug l1
Test temperature:
15 ÷ 2°C
pH:
7.5—8.1
Dissolved oxygen:
concentration of dissolved oxygen was 10.2—10.6 mg/L.
Salinity:
No data
Nominal and measured concentrations:
1, 2, 5, 10, 20 and 50 mg/L nominal.
Details on test conditions:
A 96h semi-static toxicity test was carried out with rainbow trout with daily renewal of the test solutions. Seven glass aquaria were filled with 20 litresof filtered dechlorinated mains water. Quantities of a stock solution of 2,3-epoxypropyl neodecanoate in Analar acetone were added to six of these to give an approximately logarithmic series of concentrations between 1 and 50 mg/ L. The seventh aquarium received no test substance and served as a control. Ten rainbow trout were placed in each aquarium. The fish were not fed during the test. The aquaria were gently aerated to maintain the concentrations of dissolved oxygen. At 24h intervals the number of dead fish was recorded and any dead removed, dissolved oxygen concentration and pH were measured and the test solutions were renewed.
Reference substance (positive control):
no

Results and discussion

Effect concentrations
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
ca. 5 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
No mortalities were observed at 1 and 2 mg/L. 5/10 of the fish were dead at 5 mg/L after 96 hr of treatment. Complete mortality (10/10) occured at 10, 20 and 50 mg/L. At the two highests concentration of test substance of 20 and 50 mg/L complete mortality ( 0 % survival) was observed after 24 hr of exposure.

Any other information on results incl. tables

Sublethal observations / clinical signs:
Globules of undissolved test substance, 2,3 -epoxypropyl neodecanoate were present on the surface of the water at all test concentrations. The test substance was therefore not wholly soluble at any of the concentrations tested.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The 96 hr LC50 of rainbow trout was estimated to be 5 mg/L.
Executive summary:

The acute 96 hr acute toxicity of the test substance, 2,3-epoxypropyl neodecanoate was determined by an O.E.C.D. test guideline 203 study. The 96 hr LC50 of rainbow trout was estimated to be 5 mg/L. Therefore, Neononanoic acid glycidyl ester is anticipated to have a similar rainbow trout LC50 value under the conditions of this study.
