- A stock solution with a nominal concentration of 100000 mg/l was prepared by bringing 5 grams of 4-(3-(1-naphthylamino)propyl)morpholine to a toal volume of 50 ml with dimethylformamide. Appropriate volumes of the stock solution were added to test vessels containing 15 litres of dilution water.

- A range finding test was conducted at 0 (control and solvent control), 0.10, 1.0, 10 and 20 mg/l (nominal concentrations). After 96 hours there was 100% survival in the control, solvent control and at 0.10 and 1.0 mg/l and 100% mortality at 10 and 20 mg/l.
A definitive test was condcuted at 0 (control and solvent control), 1.3, 2.2, 3.6, 6.0 and 10 mg/l (nominal concentrations). The test was terminated after 72 hours because the fish lot number could not be verfified. Thus, this test was determined to be invalid and was repeated.
A final definitive test was condcuted at 0 (control and solvent control - 0.10 ml/l dimethylformaamide), 1.3, 2.2, 3.6, 6.0 and 10 mg/l (nominal concentrations) at 22 ±1 °C. Ten fishes were added to each of the two replicate control and treatment vessels. The test was performed in 20 litre glass aquaria that contained 15 litres of solution (media depth of approximately 19 cm). Test vessels were randomly arranged in a water bath during the 96 hour test and they were loosely covered. A 16:8 hour photopriod cycle was maintained. Gentle aeration was initiated in all test vessels after 48 hours to maintain acceptable dissolved oxygen concentrations.
The number of surviving organisms and the occurence of sublethal effects (immobilization, loss of equilibrium, erratic swimming, loss of reflex, excitability, discolouration and change in behaviour) were determined visually and recorded initially and after 3, 6, 24, 48, 72 and 96 hours. Mortality observations were made twice daily and dead fishes were removed as and when observed. Parameters such as dissolved oxygen, pH, conductivity and temperature were measured and recorded daily in each test chamber. - No insoluble material was observed during the definitive test, although test vessels containing 10 mg/l 4-(3-(1-naphthylamino)propyl)morpholine at 72 hrs and all solvent control and treatment test vessels at 96 hours were observed to be slightly cloudy. No mortality was noted in the control and solvent control dose groups. At 3 and 6 hours, surviving fish exposed to 10 mg/l 4-(3-(1-naphthylamino)propyl)morpholine were lying at the bottom of the vessel and exhibited loss of equilibrium. At 24, 48 and 72 hours, fishes exposed to 6.0 mg/l were dark in colour and lethargic. At 48 and 72 hours, fishes exposed to 3.6 mg/l were dark in colour and lethargic. At 96 hours, fishes exposed to 6.0 mg/l were lethargic and lying at the bottom of the vessel. No other sublethal effects were observed during the test.
Under the conditions of the study, exposure of fathead minnows, Pimepales promelas to 4-(3-(1-naphthylamino)propyl)morpholine resulted in the following -

- The 24 and 48 hour LC50 values were calculated usng the binomical/nonlinear interpolation method and the 72 and 96 hour LC50 values were calculated using the probit method. The no observed effect concentration (NOEC) is the highest concentration of the test substance that allows at least 90% survival of exposed organismsm and does not cause sublethal effects.
