PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution containing 10 g/L was prepared using the following procedure: 10 g test item was mixed with 1.1 g NaOH (45%). 40 mL deionised water (80 – 100 °C) was added. After homogenisation with a glass stirrer, 85 mL deionised water (room temperature) was added. Then, the vessel was stirred on a magnetic stirrer. 0.5 M NaOH was added until the resulting solution was clear. The pH of the solution was adjusted to 7.8 ± 0.2 with 1 M HCl. the solution was transferred into a volumetric flask and filled up to 1000 mL with deionised water.

Additional information

Two studies investigating the toxicity to microorganism of N-methyl-N-(C18-(unsaturated)alkanoyl)glycine (EC 701-177-3) are available.

The key study was carried out according to OECD 209 under GLP-conditions with domestic activated sludge as inoculum. In this static test seven test concentrations were used (1.0, 3.2, 10, 32, 100, 320 and 1000 mg/L). The dry matter was determined as 4.2 g suspended solids/L, giving a concentration of 1.7 g suspended solids/L in the test. The highest inhibition of respiration rate was determined for the highest tested concentration (1000 mg/L nominal) with 45.3% and 43.8% inhibition. Thus, the inhibition of respiration rate is < 50%. In the study report the EC50 was extrapolated with an EC50 (3h) of 1300 mg/L. But as an extrapolation is imprecise, the EC50 (3h) > 1000 mg/L (nominal) could be stated. The NOEC (3h) was reported as 10 mg/L.

The supporting study was conducted according to OECD 209 under non-GLP conditions. Five test concentrations (1.1, 3.3, 10.7, 32.0, 100.2 mg/L) were inoculated to activated sludge. In this study, the IC50 (3h) for respiration inhibition was above the highest test concentration of 100 mg/L. A NOEC of 10 mg/L is reported.
