In the structurally related n-butyl acrylate and ethyl acrylate NOECs of 0.136 and 0.19 mg/L respectively were determined for long-term toxicity towards Daphnia magna.

Additional information

There are no data on long-term toxicity towards invertebrates for tert-Butyl acrylate. But for the structurally related ethyl acrylate and n-butyl acrylate 21-day chronic toxicity studies are available (BAMM 1997, BASF SE 2009).

 

For n-butyl acrylate there is also a chronic study available with aquatic invertebrates, performed according to OECD 211 and in compliance with GLP criteria (BASFSE, 2009). In this study, daphnids (Daphnia magna) were exposed to nominal test concentrations of 0 (control), 0.1, 0.22, 0.46, 1.0 and 2.2 mg/L under semi-static conditions. Test concentrations were analytically verified,but since the mean values of n-butyl acrylate in M4-medium were found to be in the range of 9.3 – 80.0% of the nominal concentrations, the effect concentrations were based on time-weighted mean measured concentrations. Production of young, mortality of the parent animals and young, and presence of aborted eggs were determined daily. Also, the body length of parental animals was determined at the end of exposure. No mortality was observed related to the test substance and none of the young died. Reproduction was significantly affected at 1.0 and 2.2 mg/L (nominal). Growth was significantly less in the highest tested concentration. The most sensitive effect values (measured, time-weighted averages) were concerning reproduction: NOEC (reproduction) = 0.136 mg/L; LOEC (reproduction) = 0.457 mg/L; NOEC (body length) = 0.457 mg/L; LOEC (body length) = 1.226 mg/L.

 

For ethyl acrylate a 21-day chronic study in aquatic invertebrates is available (BAMM, 1997). In this GLP-compliant study performed according to EPA OTS 797.1330, daphnids (Daphnia magna) were exposed to nominal test concentrations of 0 (control), 0.38, 0.75, 1.5, 3.0, and 6.0 mg/L for 21-days, under flow-through conditions. Test concentrations were analytically verified, but did not remain within 80 - 120% of the nominal concentrations. Therefore, the effect concentrations are based on the arithmetic means of the measured concentrations. Survival, abnormal effects, and observance of first brood of the organisms were recorded every day throughout the study. Reproductive success was measured by counting and discarding the offspring produced in each concentration every Monday, Wednesday, and Friday for the duration of the study.At termination the length of each adult daphnid was measured. Survival of Daphnia magna was shown to be significantly affected in the 0.73, 1.7, and 3.2 mg/L mean measured test concentrations. The time to first brood release, the mean young per adult reproduction day (YAD) value, body length and bodyweight was shown to be significantly affected when compared to the control for the mean measured concentration of 0.45 mg/L. Based on these results, the EC50 value (immobility) was determined to be 0.5 mg/L (measured) and the NOEC for reproduction wasdetermined to be 0.19 mg/L (measured).
