- Des segments apicaux frais ont été cultivés aérobiquement
150 ml de milieu synthétique d'eau de mer pendant 10 jours
dans les conditions suivantes selon Mendes et
Al. (2012): un Erlenmeyer de 250 ml, à pH 7,9 ± 0,3 et
salinité de 33,5 ± 0,5 ‰. Ensuite, dix segments apicaux étaient
couper en petits fragments (3-4 mm), ajouté à 150 ml de
moyen, et cultivé pendant trois jours supplémentaires. Le 13ème jour, une fois que l'algue a été acclimatée au milieu, trois à quatre
segments frais avec un poids total de 6,0 ± 0,5 mg ont été soigneusement
transféré dans 150 ml de milieu fraîchement préparé
(Mendes et al., 2012).
Afin de déterminer le temps d'exposition, le long duquel le
l'ion est en contact avec l'algue, les segments apicaux de G.
domingensis ont été exposés à Cd2 +, Cu2 + et Zn2 + de 1
à 7 jours (Tableau S2). Ces cations métalliques ont été choisis
parce qu'ils sont connus pour être toxiques pour les algues (Eklund et
Kautsky 2003). Aliquotes des solutions mères (Tableau S1)
ont été dissous dans le milieu de culture donnant l'ion final
concentrations: 0,09 mM de Cd2 +, 0,6 mM de Cu2 + et 0,8 mM
Zn2 +. Les tests toxicologiques avec non essentiel (Cd2 +, Pb2 +
et La3 +) et essentiels (Li +, Na +, K +, Sr2 +, Ca2 +, Mg2 +,
Ions Mn2 +, Co2 +, Ni2 +, Zn2 +, Cu2 + et MoO42-)
réalisée en exposant l'algue G. domingensis à leur
concentrations croissantes (tableaux S1 et S3). Les resultats
sont exprimés comme la moyenne des triplicats.

- Des cultures uniaxes de G. domingensis rouge ont été fournies par
la collection de culture d'algues, de cyanobactéries et de champignons de l'Instituto de Botânica-CCIBt, São Paulo, SP, Brésil. G.
domingensis a été maintenu dans une chambre climatique
(HotPack, USA) à 26 ± 0,5 ° C et 74 ± 10 μmol de photons
m-2 s-1 (OSRAM FL 20 W) avec un cycle lumière / obscurité de 14: 10 h.
L'irradiance a été mesurée à l'aide d'un photomètre LI-250
équipé d'un capteur quantique sphérique (LI-COR, Inc).
La température a été constamment surveillée avec
thermohydromètres (modèle 3003C). L'eau de mer synthétique
milieu a été préparé comme décrit précédemment dans
eau déminéralisée (Milli-Q System, Millipore Simplicity)
et a été stérilisé à 120 ° C pendant 35 min (Iwasaki 1961;
Mendes et al. 2012). Les vitamines ont été ajoutées après la
stérilisation par filtration à travers un millipore de 0,22 μm
Membrane PVDF. Le pH (7,9 ± 0,3) et la salinité (33,5 ±
0,5 ‰) des médias ont été surveillés à la fin de la
expérimenter avec un pH-mètre (Mettler Toledo
FE20 / EL20) et un salinomètre (Biobrix, modèle 211).

Macroalgues du genre Gracilaria ont une considérable
importance économique en tant que matière première pour la production d'agar
et appartiennent à un groupe important d'organismes qui sont
tolérant aux fortes concentrations de métal.L'inhibiteur médian
concentration (IC50) obtenues en mesurant la
rapport de la variation de masse fraîche (c'est-à-dire, les taux de croissance quotidiens) du
macroalgue Gracilaria domingensis pendant une toxicité aquatique de 48 h
les tests sont rapportés ici.L'algue a été exposée à 14
différents cations métalliques ainsi que l'anion molybdate dans
