RUN 1 – 25 mL/h: Regarding fractions 06 to 15 the solubility varied between 1.659 mg/L and 1.960 mg/L. Average solubility: 1.832 mg/L (RSD: 4.8 %).

The determined water solubilities for RUN 1 and RUN 2 show a repeatability that was significantly higher than 30 %. In addition to this the found water solubility for the lower flow rate is higher than the solubility for the higher flow rate. This is an indication that equilibrium of the aqueous phases was not yet achieved at the highest flow rate. The results for the two lower flow rates (RUN 2 and RUN 3) show a repeatability, that is < 30 %, therefore the results of these two runs are considered to be in accordance. At the end of each RUN, there was still enough test item left on the surface of the glass beads.

Conclusions:
The water solubility of 5’-O-(4,4’-Dimethoxytrityl)thymidine in double distilled water was found to be 3.0 mg/L at pH 7.1 and a temperature of 20 °C.
Executive summary:

The water solubility of 5’-O-(4,4’-Dimethoxytrityl)thymidine was determined in double distilled water according to the "column elution method" described in the European Commission Council regulation (EC) No 440/2008, Annex, Part A, A.6., OECD-guideline 105 and the EPA Product Properties Test Guideline OCSPP 830.7840 and found to be 3.0 mg/L at pH 7.1 and a temperature of 20 °C.
