A limit test with a nominal test substance concentration of 100 mg per litre dilution water was performed
- Sampling method: Samples of the test medium for determination of the actual test substance concentration were taken immediately before introduction of the fish and approximately after 4, 24, 48, 72 and 96 hours. One sample each was taken and then analysed in duplicate by HPLC.
When the test media were changed after 24, 48 and 72 hours of exposure, samples were taken from the old as well as from the new media.
- Sample storage conditions before analysis: The samples were deep frozen on the day of sampling and unfrozen for the analysis.

PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: A solution with a nominal test substance concentration of 100 mg/L was prepared by dissolving 1031 mg test substance (solution as obtained by the sponsor) in 5 litres
dilution water and manually homogenization. In order to maintain the actual test substance concentration above 80 % of the actual starting
concentration new test media were prepared for test media change at 24, 48 and 72 hours after the start of the test.
- Controls: Control animals were holded in dilution water.
- Evidence of undissolved material: No: Visual observations were made at the same time as the animal observation and the media changes.
The control medium and the test media were clear and colourless throughout the test period.

According to the EC directive, a limit test may be performed at 100 mg/L in order to demonstrate that the LC50 is higher than this concentration.
Therefore it was decided to perform a limit test with only one concentration (100 mg/L).

Executive summary:

A limit-test with a concentration of nominal 100 mg test substance per L dilution water was performed.

7 Zebrafish(Danio rerio) were exposed in the test substance group and 7 in the control group. Their length was between 3.1 and 3.5 cm.

A ca. 50 % solution of test substance in water was used. According to the sponsor the actual concentration of the test substance was 48.5 % w/w. A nominal test substance concentration of 100 mg/L was therefore prepared by dissolving 1031mg test substance in 5 litres dilution water and manual homogenization. The dilution water was local tap water, mixed 1:1 with deionised water, to achieve the appropriate water hardness as required by the guideline.

This dilution water was also used as the negative control substance and for fish holding prior the test.

The exposure of the Zebrafish was for 96 h in a semi-static test with renewal of test and control media after 24, 48 and 72 hours.

The results of the study revealed a LC50 after 96 h of exposure of more than 100 mg/L, a NOEC of 100 mg/L and a LC0 after 96 hours greater or equal 100 mg/L.
