Ironpentacarbonyl, Fe(CO)5, is severly toxic upon inhalation (estimated LC50 ca. 10 ppm; 81 mg/m3). In a 28 days inhalation study 1 ppm still showed a slight increase in lung weights whereas 3 ppm caused severe lung damage and partial lethality and 0.3 ppm were without effect. The dose effect relation appears to be steep. This type of dose-effect and dose-response relation does not indicate major intraspecies differences. However, it is not excluded that 1 ppm and lower concentrations may accumulate more iron upon longer exposure. In the absence of a 90-day study or a bioaccumulation study, this potential of cumulative damage should be treated with a safety factor of 10 upon the LOAEL. An allometric factor for the cross-species extrapolation is not necessary due to the inhalation route in the experiment. The resulting overall assessment factor of 10 indicates a chronic inhalation DNEL of 0.1 ppm (0.81 mg/m3). The German MAK level is also 0.1 ppm. A 2-fold higher DNEL is proposed for a single short exposure period.

The lungs appeared to be the most sensitive target organ. On the other hand, also systemic toxicity has been observed at the higher exposure levels. Therefore the same DNELs are applied for systemic and local toxicity and no further discrimination is made between both DNELs.

No DNELs were derived for dermal exposure due to the high systemic toxicity from the dermal route.
