PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The test substance is a poorly water soluble mixture. Therefore a water accommodated fraction (WAF) was prepared using a slow-stir liquid-liquid saturator technique [Ref. 9, 10]. The required volume of test media was added into a glass aspirator bottle (approx. 20 L) with a bottom side-outlet attached to a stopcock. The necessary volume of the test substance (2.67 mL) was calculated from the density and was pipetted carefully on the water surface. The bottle was closed and the solution was stirred on a magnetic plate for about 1 day. The stirring is slow (approx. 100 rpm) so that no vortex formed. The required volume (10 L) of the WAF was removed from the bottom of the bottle and visually inspected for the presence of any undissolved test substance. The control was prepared in the same way but without test substance and in closed 10 L bottles. The control preparation was used directly in this bottles.

Preparation was performed in closed vessels with a tap. The headspace was reduced to a minimum. Individual loading rate of 100 mg/l was prepared. A 1-day period of magnetic stirring was applied. During the stirring period care was taken to prevent the formation of an emulsion. Therefore, the vortex was in all cases less than 10% of the height of the medium, preventing the test substance being pulled down and hitting the stirring bar. The vortex was similar in all vessels. At the end of the stirring a floating layer and droplets of the test substance suspended in the solution were observed in the vessels. After a 1-hour settlement period the aqueous WAFs were collected using a tap at the bottom of the vessel and used as test concentrations (a small volume of the WAF used to flush the tap was discarded). A macroscopic observation of the final test solutions revealed that they were all colorless and free of undissolved material.

Individual treatment solutions were prepared by adding the test substance, via syringe, to 17 L of laboratory blend water in 20 L glass carboys that were sealed to minimize volatile loss of test material. The stock solutions were mixed for 24 hours with a vortex of <10% (1-2 cm vortex). Mixing was performed using a magnetic stir plate and teflon stir bar. After mixing, the solutions were allowed to settle for one hour and the Water Accommodated Fraction (WAF) was then siphoned from the bottom of the mixing vessel and added to the test vessels.
