Description of key information
Acute oral toxicity:
The study was designed and conducted to determine the acute oral toxicity profile of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl] trimethylammonium chloride in Sprague Dawley rats. Initially, three female animals were treated at the dose level of 300 mg/kg body weight of the test item (Step - I). Administration of the test item at 300 mg/kg did not result in any signs of toxicity and mortality at 24 hours after the dosing. As no mortality was observed at 24 hours after the dosing, three female animals were added to the study and treated with the same dose of 300 mg/kg of the test item (Step - II). Administration of the test item at 300 mg/kg did not result in any signs of toxicity and mortality after the dosing. No mortality was observed at 300 mg/kg dose group, hence additional three female animals were treated with the higher dose of 2000 mg/kg of the test item (Step - I). Administration of the test item at 2000 mg/kg did not result in any signs of toxicity and mortality after the dosing. As no mortality were observed at 24 hours after the dosing, hence additional three female animals were treated with the higher dose of 2000 mg/kg of the test item (Step - II). Administration of the test item at 2000 mg/kg did not result in any signs of toxicity and mortality after the dosing. Gross pathological examination did not reveal any abnormalities in animals from 300 mg/kg and 2000 mg/kg dose groups. It was concluded that the acute oral median lethal dose (LD50) of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl]trimethylammonium chloride supplied by Sustainability Support Services (Europe) AB, Sweden, when administered to Sprague Dawley rats was found to be 2000 mg/kg body weight. Thus by considering the CLP criteria for acute toxicity rating for the chemicals, it infers that [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl]trimethyl ammonium chloride falls into the “Category 4” which is relatively nontoxic.
Acute Dermal toxicity:
The study was designed and conducted to determine the acute dermal toxicity profile of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl] trimethylammonium chloride in Sprague Dawley rats. The test item was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days.Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment. It was concluded that the acute dermal median lethal dose (LD50) of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl] trimethylammonium chloride supplied bySustainability Support Services (Europe) AB, Sweden, when administered to male and female Sprague Dawley rats was found to be greater than 2000 mg/kg body weight. Thus by considering the CLP criteria for acute toxicity rating for the chemicals, it infers that [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy -2-naphthyl] trimethylamm onium chloride does not exhibits acute toxicity by the dermal route.

Acute oral Toxicity:
Various data available for the target chemical was reviewed to determine the oral toxic nature of Basic Brown 17 (IUPAC name:
[8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl] trimethylammonium chloride). The studies are as mentioned below:
The study was designed and conducted (Sustainability Support Services, 2016) to determine the acute oral toxicity profile of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl] trimethylammonium chloride in Sprague Dawley rats. Initially, three female animals were treated at the dose level of 300 mg/kg body weight of the test item (Step - I). Administration of the test item at 300 mg/kg did not result in any signs of toxicity and mortality at 24 hours after the dosing. As no mortality was observed at 24 hours after the dosing, three female animals were added to the study and treated with the same dose of 300 mg/kg of the test item (Step - II). Administration of the test item at 300 mg/kg did not result in any signs of toxicity and mortality after the dosing. No mortality was observed at 300 mg/kg dose group, hence additional three female animals were treated with the higher dose of 2000 mg/kg of the test item (Step - I). Administration of the test item at 2000 mg/kg did not result in any signs of toxicity and mortality after the dosing. As no mortality were observed at 24 hours after the dosing, hence additional three female animals were treated with the higher dose of 2000 mg/kg of the test item (Step - II). Administration of the test item at 2000 mg/kg did not result in any signs of toxicity and mortality after the dosing. Gross pathological examination did not reveal any abnormalities in animals from 300 mg/kg and 2000 mg/kg dose groups. It was concluded that the acute oral median lethal dose (LD50) of [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl]trimethylammonium chloride supplied by Sustainability Support Services (Europe) AB, Sweden, when administered to Sprague Dawley rats was found to be 2000 mg/kg body weight. Thus by considering the CLP criteria for acute toxicity rating for the chemicals, it infers that [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2-naphthyl]trimethyl ammonium chloride falls into the “Category 4” which is relatively nontoxic.
In another study for the target chemical by Scientific Committee on Consumer Safety (2003), CFY rats (male/female) were exposed orally to the test chemical (8-((4-Amino-3-nitrophenyl)azo)-7-hydroxy-2-naphthyl)trimethylammonium chloride. The rats (CFY strain) were treated with Arian or Sienna Brown with a range of dose levels from 0.1 to 16 g/kg body weight. The test compound was prepared as 10 and 40 % (w/v) suspensions in 1% aqueous methylcellulose. The test chemical conc. used for the study was 0, 100, 1000, 4000, 8000 and 16000 mg/kg bw, respectively. Rats dosed with the vehicle alone served as controls. Signs or reaction to treatment, observed shortly after dosing, included lethargy, piloerection, decreased respiratory rate and abnormal body carriage (hunched posture). After oral administration of the test substance, mortality of 2 male rats and 1 female rat from the highest dose (16000 mg/Kg bw) group were observed. After 14 days of observation, the LD50value was determined to be between 8000 and 16000 mg/kg bw, respectively.
In the above mentioned study by Scientific Committee on Consumer Safety (2003), CF1 male mouse were exposed orally to the test chemical (8-((4-Amino-3-nitrophenyl)azo)-7-hydroxy-2-naphthyl)trimethylammonium chloride. The test chemical conc. used for the study was 1000, 2510 and 5010 mg/kg bw, respectively. The test material was given to CF1 mice once by gavage at three dose levels between 1.00 and 5.01 g/kg body weight and administered at a volume of 0.2 ml/10 g body weight. All animals were observed for a period of 7 days. After oral administration of the test substance, no mortalities were recorded during observation period of 7 days. The LD50value was determined to be greater than 5000 mg/kg bw.
