- - Cell shape: No morphological abnormalities were observed in the control and in the concentrations of 6.25, 12.5, 25.0 and 50.0 mg/L during the experiment. In the highest test item group (100.0 mg/L) thin cells were observed together with normal cells at 48 and 72 hours.

The effect of the test item on the growth of a unicellular green algal species Raphidocelis subcapitata was determined according to OECD 201, Regulation (EC) No. 226/2016 Method C.3 and EPA OPPTS 850.5400 and under GLP. The study was conducted under static conditions with an initial cell density of 1E+4 cells/mL for a period of 72 hours. Five test concentrations - 6.25, 12.5, 25, 50 amd 100 mg/L, and control were tested. The measured concentrations of the test item at the start and at the end of the test were in the range of 99 - 100 % of the nominal concentration. The measured concentrations deviated not more than 20 % from the nominal concentration, therefore, the nominal concentrations were used to determine exposure concentrations. The EC50-values for inhibition of growth rate (ErC50) and for inhibition of yield (EyC50) were >100 mg/L. The ErC10 was determined to be >100 mg/L. The EyC50 was 66.72 mg/L and the NOEC was determined to be 100 mg/L. The test item showed no toxic effects on the growth of the Raphidocelis subcapitat up to the concentration of 100 mg/L.
Reference
Pre-experiments
In the preliminary tests, the test item showed 15 % growth inhibition at 100 mg/L tets concentration when the pH of the test item solution was not adjusted. After adjustment of the pH, the inhibition at 100 mg/L concentration level was 9.93 %. In the second preliminary test, performed with adjusted pH, the inhibition of the algae was at a maximum of 4.99 % at treatment level of 100 mg/L.
Validity criteria
- Cell density in the control cultures increased by a factor of more than 16 (by a factor of 50.50) within 72 hours.
- The mean coefficient of variation for section-by-section specific growth rates (days 0-1, 1-2 and 2-3, for 72 h-tests) in the control cultures did not exceed 35 %.
