In accordance with Regulation (EC) No 1907/2006 Annex XI, 1.5 a read across to a structurally related substance Fatty acids, C16-18 and C18-unsatd., hexaesters with dipentaerythritol (CAS 68604-38-6) was conducted to fulfill the data requirements according to Annex VII-IX of Fatty acids, tall oil, esters with dipentaerythritol (CAS 70913-98-3) regarding long-term toxicity to aquatic invertebrates. This read-across is justified in detail in the overall summary (IUCLID Section 6.1) and within the analogue justification in IUCLID Section 13. The fatty acid chain lengths of C16, C18 and C18 unsatd. of the read-across substance is identical to those of the target substance. In addition, the read-across substance contains the same alcohol component, dipentaerythritol, and is a hexa-ester.
The chronic toxicity of the read-across substance Fatty acids, C16-18 and C18-unsatd., hexaesters with dipentaerythritol (CAS 68604-38-6) to aquatic invertebrates was tested in a GLP guideline study following OECD 211 (Caduff, 2013). Daphnia magna was exposed to the test substance under semi-static test conditions. The test solutions were prepared as Water accommodated fractions (WAF). During the exposure period of 21 days the mortality and reproduction rate of the test animals was recorded. The substance affected neither the survival rate nor the reproduction rate of Daphnia magna. Thus, a NOELR (21 d) ≥ 100 mg/L was derived.
Based on this result from a structurally related read-across substance (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which is characterized by a similar ecotoxicological profile and a similar structure, it can be concluded that Fatty acids, tall oil, esters with dipentaerythritol (CAS 70913-98-3) will not exhibit chronic effects to daphnids up to the limit of water solubility.
