Description of key information

Two short-term toxicity tests with orange killifish (Oryzias latipes) exposed to 2-hydroxyethyl methacrylate and conducted under GLPs, under sem-static or flow-through conditions and following standard test guidelines (OECD 203 and OECD 204) resulted in both a 96-hour and 14-day LC50 value of >100 mg/L.

Five acute freshwater fish studies were available for 2-hydroxyethyl methacrylate with three considered reliable for assessment of this endpoint. Two studies were conducted with the orange killifish (Oryzias latipes), while the other study was conducted with the fathead minnow (Primephales promelas). These studies were found to be of good quality and assigned a Klimisch score of one or two. The two orange killifish studies were conducted following standard OECD guidelines and under GLPs. One study was conducted under semi-static conditions for 96 hours and the second study conducted under flow-through conditions for 14-days. The fathead minnow study was conducted under flow-through conditions for 96 hours and followed ASTM guidelines but not under GLPs. The study was conducted under flow-through conditions for 96 hours. The studies concluded that the 96-hour and 14-day LC50 values for the ricefish were both >100 mg/L and the 96-hour LC50 for the fathead minnow was 227 mg/L. Since these toxicity results were from studies with different fish species and/or varying exposure periods, the lowest of the three values (>100 mg/L) was selected as the key parameter for this endpoint. 

The remaining two fish studies (supporting studies) with the golden ide (Leuciscus idus melanotus) and the goldfish (Carassius auratus) were not considered reliable for assessment. The studies were conducted under static conditions and did not follow standard test guidelines or GLPs.  The golden ide study reported a 48-hour LC50 value of 360 mg/L and the goldfish study reported a 72-hour LC50 value of 374.5 mg/L, respectively.
