Description of key information
The 96h-LC50 of 1.3 mg/L (nominal test material concentration) was determined for N-[2-hydroxy-3-(C12-16-alkyloxy)propyl]-N-methyl glycinate (OECD 203, reliability 2). The results of this study have been read-across to nonylphenol ethoxylate, sarcosine derivative.

The acute toxicity to fish was determined for N-[2-hydroxy-3-(C12-16-alkyloxy)propyl]-N-methyl glycinate. The results of this study have been read-across to nonylphenol ethoxylate, sarcosine derivative. The two substances have a similar chemical structure in the 2-hydroxy propyl-N-methyl glycinate moiety, but they differ in the alkyloxy moiety attached to the propanol structure. Where the test substance has a straight C12-16 chain attached to the propanol via an ether link, the registered substance has 4-nonylphenol ethoxylate linked to the propanol via the ether bonding. The effects of the different alkyloxy moieties on the acute toxicity to fish are unknown. The structural formulas of the test substance and registered substance are depicted in the attached document.
The 96h-LC50 for N-[2-hydroxy-3-(C12-16-alkyloxy)propyl]-N-methyl glycinate of 1.3 mg/L (test material) was determined in an acute toxicity test with rainbow trout (Oncorhynchus mykiss) according to OECD TG 203 under GLP conditions. The study was assigned reliability 2 due to the lack of a certificate of analysis and as chemical analyses were not sufficiently reliable, nominal exposure concentrations of the test material have been used. Information on active ingredient content of the test material is lacking. Therefore LC50 is based on the nominal test material concentration.
The acute toxicity test with fish was performed according to OECD TG 203 under GLP.The study was performed to assess the acute toxicity of the test material, DS 100, to rainbow trout (Oncorhynchus mykiss). The method followed that described in the OECD Guidelines. Following a preliminary range-finding study, fish were exposed, in groups of 10, to an aqueous dispersion of the test material over a range of concentrations of 1.0, 1.8, 3.2, 5.6 and 10 mg/l for a period of 96 hours under semi-static test conditions. The number of mortalities and any sub-lethal effects of exposure in each test and control vessel were determined 3 and 6 hours after the start of exposure and then daily throughout the study until termination after 96 hours.
The 96-Hour LC50, based on nominal test concentrations was 1.3 mg/l with 95% confidence limits of 1.0 - 1.8 mg/l. The No Observed Effect Concentration was 1.0 mg/l. Analysis of the test solutions at 0, 24 and 96 hours showed the measured concentrations to be near nominal with the exception of the 1.0, 1.8 and 3.2 mg/l test groups at 24 hours which gave measured values of 79, 78 and 79% of nominal respectively. Given that these results were only 1 - 2% less than the required 80% of nominal value and all other results were near nominal it was considered justifiable to base the LC, values on nominal test concentrations only.
